Quantifying Bisphenol from Food Packaging and Assessment of its Cytotoxic Potential

Abstract

Bisphenol A (BPA) is a compound that is released from PET packaging during storage bottled water and can have a harmful effect on human health. This study aimed to investigate the cytotoxic potential of BPA at the Caco-2 cell line. Bottled water samples coming from two companies were analyzed starting from 1 month to 12 months of storage Quantification of BPA in water stored in PET containers was carried out by a spectrometric method. Calibration was performed for BPA concentrations ranging between 0.01 and 0.09 mg/L. Caco-2 cell cultures were treated for 24 hours with different BPA concentrations (from 0.1 mM to 1 mM). Viability and cell cytotoxicity analysis was performed by MTT assay. BPA concentration in one water sample ranged from 0.09 to 0.92 mg/L while for second water sample BPA concentration ranged from 0.03 to 0.73 mg/L. After 10 months from packing, BPA concentration began to exceed the 0.6 mg/L limit stated by Regulation (EU) no. 10/2011. Cell viability decreased in a dose dependent manner and reached the minimum of 3.8% for the 1 mM BPA concentration.
