Abstract

Validity and correctness test verification of the measuring software has been a thorny issue hindering the development of Gear Measuring Instrument (GMI). The main reason is that the software itself is difficult to separate from the rest of the measurement system for independent evaluation. This paper presents a Virtual Gear Measuring Instrument (VGMI) to independently validate the measuring software. The triangular patch model with accurately controlled precision was taken as the virtual workpiece and a universal collision detection model was established. The whole process simulation of workpiece measurement is implemented by VGMI replacing GMI and the measuring software is tested in the proposed virtual environment. Taking involute profile measurement procedure as an example, the validity of the software is evaluated based on the simulation results; meanwhile, experiments using the same measuring software are carried out on the involute master in a GMI. The experiment results indicate a consistency of tooth profile deviation and calibration results, thus verifying the accuracy of gear measuring system which includes the measurement procedures. It is shown that the VGMI presented can be applied in the validation of measuring software, providing a new ideal platform for testing of complex workpiece-measuring software without calibrated artifacts.

Abstract

Resources and the environment have always been the two important natural factors that affect people’s lives. In recent years, the problem of resources and the environment has increasingly become an important issue that people are concerned about. This study discusses the use and consumption of energy and the impact of environmental pollution on economic development under sustainable economic development. This paper takes Panzhihua as an example to analyze the impact of energy and environment on the economy, and proposes solutions to improve economic development, which is of strategic significance for the future development of Panzhihua City. In this paper, the system dynamics method is used to decompose the Panzhihua large-scale system into three parts and carry out modeling and simulation to explore the connection between them. Based on the data from 2007 to 2015 in Panzhihua City, simulations have been carried out to obtain qualitative and quantitative analysis of certain simulation curves of the energy-environment-economy 3E system (hereinafter referred to as 3E system) from 2007 to 2030 to ascertain the future development pattern of Panzhihua City. The results show that when the 3E system is a coordinated development model, economic development and environmental protection have a good development trend at the same time, which is applicable to the future development of Panzhihua City. This model has good reference suggestions and application prospects for urban development. We want to give Panzhihua City the following suggestions: (1) Continue to focus on the secondary industry and increase competitiveness. (2) Increase the investment funds in environmental protection and achieve sustainable economic development.

Abstract

The fatigue problem induced by wave loads and floater movements of dynamic umbilicals in deepwater is studied. The prediction method of the fatigue life is investigated by considering no-linear local stress due to contact and friction between components of the umbilical. A case study of a dynamic umbilical for 1500 meters depth in South China Sea was presented. The results showed that the more accurate result of the fatigue life was calculated by considering non-linear local stress. The fatigue life by considering local stress with the no-slip assumption was conservative. An obviously longer fatigue life was obtained by considering local stress with the full-slip assumption.

Abstract

Background: Ovarian carcinoma represents one of the most insidious and aggressive cancers. Ovarian carcinoma is the most lethal gynecological malignancy. To discover new relevant biomarkers to increase specificity and sensitivity for early diagnosis and prognosis of ovarian cancer is important and urgently needed. FOXO3a possesses a large series function including cellular proliferation, transformation, differentiation, and longevity. Jab1 is also known as subunit 5 (CSN5) of the COP9 signalosome (CSN), a multifunctional protein complex involved in modulating signal transduction, gene transcription, and protein stability.

Objective: To investigate the importance of FOXO3a and Jab1 in ovarian cancer.

Method: Immunohistochemical analysis was performed on formalin-fixed paraffin sections of 46 specimens. Statistical analysis was conducted using the Stat View 5.0 software package.

Result: We found that Foxo3a expression correlates significantly with FIGO disease stage (p < 0.05) and lymph node involvement. Jab1 expression correlates significantly with disease stage and lymph node involvement (p < 0.05). A multivariate Cox proportional hazard model showed that Foxo3a and Jab1 were the strongest independent predictors of survival (p < 0.05), the second predictor being FIGO disease stage and lymph node involvement.

Conclusion: Understanding the precise role of Jab1 and Foxo3a in ovarian cancer progression will not only increase our knowledge of the biology of ovarian cancer but may also enable development of an novel therapeutic strategy via suppression of Jab1.

Abstract

Background: Cyclin-dependent kinase inhibitor p27Kip1 is a new class of tumor suppressor in a dosage dependent manner to control cell cycle progression. Human epididymis protein 4 (HE4) is a potentially useful biomarker for ovarian carcinoma, comparable with cancer antigen 125 in identifying women with ovarian cancer, both localized and advanced. However, the prognostic significance of p27Kip1 and HE4 in ovarian cancer is unclear.

Objective: Investigate the expression of p27Kip1 and HE4 in the progression of human ovarian cancer.

Material and method: Immunohistochemical analysis was performed on formalin-fixed paraffin sections of 61 specimens. The association between HE4 and p27Kip1expression and clinicopathological features was analyzed using χ2-test. For analysis of survival data, Kaplan-Meier curves were constructed, and the log-rank test was performed.

Results: The expression of p27Kip1 negatively related with HE4 expression, but it correlated significantly with lymph nodes. On the other hand, HE4 expression correlated significantly with disease stage and lymph nodes. Kaplan-Meier analysis of the survival curves of p27Kip1 and HE4 revealed a highly significant separation between low vs. high expressers in ovarian carcinoma.

Conclusion: Expression of HE4 was up-regulated significantly in human ovarian carcinoma. Overexpression of HE4 might be responsible for oncogenesis and development of ovarian carcinoma. HE4 and p27Kip1 may be of prognostic significance in human ovarian cancer.

Abstract

Optically stimulated luminescence (OSL) dating was applied to sediment samples in order to provide age constraints on the palaeo-earthquake events that are recorded in a trench on the Lingwu fault near Yinchuan on the northeastern margin of the Tibetan Plateau, China. Combining seventeen OSL ages with field observations in the trench shows that at least four events took place at this specific site, and that this fault has been active over the past 200 ka.

Abstract

African swine fever is an acute, febrile and highly virulent porcine disease causing serious economic losses worldwide. The pK205R protein of the African swine fever virus (ASFV) is largely expressed in the early stages of infection, which has given the K205R gene extensive attention. In this study, the ASFV K205R was cloned and expressed in Escherichia coli BL21 (DE3). Expression of histidine-tagged pK205R with a molecular mass of 44 kDa was determined by 12% sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot analysis. Optimisation of culture conditions allowed induction of the recombinant protein with 0.4 mM Isopropyl β-D-thiogalactoside (IPTG) at 37°C for 2 h. The protein existed in cellular supernatant and was purified using a Ni-NTA resin column. The purified protein was used to immunize rabbits four times to enable the production of polyclonal antibodies, and the antiserum titre was detected by ELISA. The results showed that the purified pK205R can react with ASFV positive serum specifically by Western blotting. The pK205R had high antigenicity, which indicated that pK205R could be used as an antigen for detection of ASFV-specific antibodies in ELISA testing, and the recombinant protein could contribute to further research of the action and structure of pK205R.

Summary

Background: The aim of this study was to investigate the role of pulmonary surfactant-associated protein B (SP-B) expression in the pathogenesis of neonatal respiratory distress syndrome (RDS) via detecting the protein and mRNA expression of SP-B.

Methods: A total of 60 unrelated neonates who died of RDS were chosen as the RDS group and then subgrouped into ≤32 weeks group, 32∼37 weeks group and ≥37 weeks group (n=20). Sixty neonates who died of other diseases were enrolled as controls and subdivided into 3 matched groups based on the gestational age. Western blot assay and RT-PCR were employed.

Results: In the RDS group, SP-B protein expression was reduced or deficient in 8 neonates of which 6 had no SP-B protein expression. In the control group, only 1 had reduced SP-B protein expression. The reduced or deficient SP-B protein expression in 9 neonates of both groups was noted in the ≥37 weeks group. In the RDS group, the SP-B mRNA expression was significantly lower than that in the control group. In the ≤37 weeks group, SP-B mRNA expression was comparable between the RDS group and control group. In the 32∼37 weeks group, the SP-B mRNA expression in the RDS group was significantly reduced when compared with the control group. In the ≥37 weeks group, the SP-B mRNA expression in the RDS group was dramatically lower than that in the control group.

Conclusions: Alteration of SP-B expression is present at transcriptional and translational levels. Reduction of SP-B mRNA and protein expression is involved in the pathogenesis of RDS.

Abstract

Background: CA16 and enterovirus 71 are two key etiological agents for children’s hand-foot-mouth disease (HFMD). Large-scale HFMD outbreaks have taken place every year in Liaocheng City, Shandong Province of China since 2008.

Objective: We investigated the genetic background and phyletic evolution of coxsackie virus A16 (CA16)-related severe HFMD in children from Liaocheng City.

Method: CA16 was screened from throat swab and anus specimens obtained from children with severe HFMD between 2008 and 2010. Specific primers were used to amplify the VP1 region of CA 16 for sequence analysis.

Result: A total of 461 specimens were detected to be enterovirus positive from 2008 to 2010 and 401 specimens were CA16 positive. The nucleotide and amino acid sequences of 16 isolates from children with severe HFMD were compared with the reference sequences, and the nucleotide homology was 91.43%-98.65%, and the amino acid homology was 97.98%-100%. Of the16 isolates, 9 isolates and BJ03-ZDP (AY821798), Shzh00-1 (AY790926), Shzh05-1 (EU262658), and GZ08 (FJ198212) strains isolated from Chinese mainland were located on the same branch; the remaining 7 isolates, the strains isolated from Malaysia, Thailand, Vietnam, Australia and other neighboring countries. AF177911 from Taiwan andshzh01-69 strain (AY895111) from Shenzhen were located on another branch.

Conclusion: CA16 is one of the major pathogens of HFMD and the homology of strains is high.
